Human Plasma Protein Binding

 

 

Description

The purpose of this assay was to measure protein binding (%PB) of test compounds in human serum. Approximately 10 µL of 10mM test compound in DMSO was used to determine the %PB value. 

 

Method

Two assay procedures (ultracentrifugation and equilibrium dialysis) are provided. Use the protein binding assay appropriate to your needs, based on the following considerations. 

 

The ultrafiltration protein binding assay may result in high non-specific binding, but the addition of binding inhibitors as indicated in the protocol can help to alleviate this drawback. 

 

The equilibrium dialysis protein binding assay is appropriate for compounds which show high non-specific binding (NSB>50%).
